The father-of-three, who is currently in a rehabilitation clinic dealing with 'personal issues', was said to have known about the woman's pregnancy, but did not tell his actress wife.
A source close to the 40-year-old told RadarOnline.com: 'Jesse got a mistress pregnant. Jesse knew about it.'
The source added the woman did not give birth.
The unnamed woman was one of four mistresses, including tattooed model Michelle 'Bombshell' McGee, James was alleged to have affairs with during his nearly five year marriage to Bullock.
The Oscar winner has been in hiding - only being photographed once covering her face with a large sun hat on Wednesday - since news of James' alleged infidelities broke on March 17.
Bullock, 45, was seen hiding from a huge throng of paparazzi as she visited her 'dear friend' Gabriel Brener, at his Beverly Hills home.

'The single most important thing to Mr. James and the children is that the marriage somehow survive.'
Friends of the actress said she is still in shock over the revelations and is undecided over her next move.
A friend told People magazine: 'She's in shock. She's sad - but getting angrier.
'They're in a celebrity marriage where you really have to trust each other because the opportunities to stray are huge. He was a champ around her. He would make her feel like a million bucks.'
Comedian George Lopez, whose TV show is produced by Bullock, said he briefly visited the actress on Saturday night.

His spokesperson said a statement that James 'realised that this time was crucial to help himself, help his family and help save his marriage.'
The type of treatment facility was not specified.
Bullock was on a career high that culminated with a Best Actress Oscar win for The Blind Side just 10 days before rumours of James' infidelity surfaced.
The two walked hand-in-hand down Oscars red carpet, celebrated together at the parties afterward and were photographed outside a Long Beach burger joint the day after the awards.
She moved out of the marital home on March 15, two days before his alleged affair with McGee became public.
